Abstract
Ibuprofen is one of the drugs practically insoluble in water. Ibuprofen was made Orally Disintegrating Tablets dosage form to improve the solubility of ibuprofen to get a good bioavaibility. Orally Disintegrating Tablets (ODT) is a common abbreviation for a quick disintegrated tablet in the oral cavity and the particles are ingested substances showed immediate release. The tablet is intended to rapidly disintegrate in the mouth when contact with saliva in less than 60 seconds or preferably less than 40 seconds (Kundu & Sahoo 2008). Additional material filler, binder and disintegrater greatly affect drug release. This study was aim to determine the different proportion of combination excipients mannitol and binder microcrystalline cellulose to get the optimum formula by Simplex Lattice Design method. Ibuprofen Orally Disintegrating Tablets were made by dry granulation method. Based on the Simplex Lattice Design method, it was made by using three formulas with the proportion between mannitol and microcrystalline cellulose as F1 (100%: 0%), F2 (50%: 50%), F3 (0%: 100%). Each formula was made into granules and tablets, then tested against to critical point parameters are optimized using Design Expert v7.1.5 program which includes the flow of time, water absorption, disintegration time and dissolution. Optimum formula then validated by comparing the t test (t-test) 95% significance level. The results showed no difference between the prediction and experiment with methods of Simplex Lattice Design obtained optimum formula of 67% mannitol and microcrystalline cellulose by 33%.
