Abstract
Blackcumin (Nigella sativa) seed contains polyphenol, essential oil, saponin, alkaloid and tannin. While phaleria leaf contains alkaloid, flavonoid, polyphenol and saponinwhich efficacious as antiasthma. This study was aimed to determine the antiasthmaeffect in administration of single and combinationextracts of blackcuminseeds and phalerialeaves by counting parameter of mast cell granulenumber in Balb/C mice bronchial and to determine more effective concentrations between Nigella sativa L.seeds and Phaleria macrocarpaleaves or bothcombination. Test animals were divided 7 treatment groups, consisting of 5 mice: group I positive control 0.0026 mg/20 g BWmice. Group II negative control 0.5% CMC. Group III combination ofblackcuminand phaleria50%:50%, group IV 75%:25%, group V 25%:75%, group VI blackcumin100%, group VII mahkota dewaleaves 100%. All groups were sensitized by 2.5 mg OVA, dissolved by 7.75 ml Al(OH)3 0 and 14thdays. Provision of extract, Profilas® and CMC were conducted 1stto27thdays. 28thdays bronchial tissue was made immunohistologypreparate and stained by methylene blue. Observed with light microscope 100xmagnification. Results obtained were analyzed by Mann Whitney and Krusskal Wallis tests. Conclusion, provision combinationextractof 96% ethanol blackcuminseeds and phalerialeaves with doses of 37.5 mg/20 g BW mice and 12.5 mg/20 g BW mice had the anti-asthmatic effect with count reduced of mast cell granules number in Babl/C mice bronchial with average number mast cells was 60 cell mastgranules.
