Abstract

Wangon leaves (Olax scandens Roxb.) contain chemical compounds such as flavonoids, steroids, saponins, and tannins. In the previous study had the efficacy as an antibacterial using n-hexane and methanol solvents with cold percolation method. This study aimed to determine the antibacterial activity of ethanol extract, fraction of n-hexane, ethyl acetate, and water from wangon leaves (Olax scandens Roxb.) to the growth of Escherichia coli ATCC 25922. Wangon leaf powder was extracted by maceration method using 96% ethanol. The extract obtained was fractionated by using n-hexane, ethyl acetate, and water solvents. The fractination results were tested for antibacterial activity againts Escherichia coli ATCC 25922 using a diffusion method with a concentration of 40%., 20%, 10%, 5% to find out the most active fraction. The most active fraction from diffusion is then followed by a dilution test with concentrations starting at 40%, 20%, 10%, 5%, 2,5%,1,25%, 0,625%, 0,3125%, 0,156%, 0,078% to determine the Minimum Barrier Concentration (MBC) and Minimum Kill Concentration (MIC). The results showed that ethanol extract, fraction n-hexane, ethyl acetate, and water from wangon leaves (Olax scandens Roxb.) had antibacterial activity againts Escherichia coli ATCC 25922. The most active fraction obtained was ethyl acetate fraction with a concentration of 40% which had a inibition zone of 10,6 mm. The results of the Minimum Kill Concentration (MIC) were obtained at a concentration of 40% and the Minimum Barrier Concentration could not be known.
