Abstract

BUDI UNIVERSITY, SURAKARTA. Karandas leaves (Carissa carandas L.) is one of the plants that has many properties in traditional medicine such as wound, diarrhea and syphilis. This research aims to determine the antibacterial activity of ethanol extract, n-hexane fraction, chloroform fraction, and water fraction of karandas leaves, to find out the most active fraction, and to know the Minimum Inhibitory concentration and Minimum Killing Concentration of the most active fraction of Staphylococcus aureus ATCC 25923. Karandas leaves extracted by maceration method with ethanol solvent 70%. The extracts obtained were fractionated with n-hexane, chloroform and water solvents. Results of extraction and fractionation tested antibacterial activity against Staphylococcus aureus ATCC 25923 with diffusion method at concentrations of 50%; 25%; 12,5% and method dilution at concentrations of 25%; 12,5%; 6,25%; 3,12%; 1,56%; 0,78%; 0,39% and 0,195%. Positive control used Siprofloksasin and negative control used DMSO 5%. The identification of chemical content is done by color reagent and Thin Layer Chromatography (TLC). Research results were analyzed with statistical test Analysis Of Variants (ANOVA) One Way. The results of the study showed ethanol extract 70%, n-hexane fraction, chloroform fraction, and water fraction of karandas leaves has antibacterial activity against Staphylococcus aureus ATCC 25923. Chloroform fraction with a concentration of 50% was the most active fraction with a barrier diameter of 15.00 mm and a Minimum Killing Concentration is 6,25% against Staphylococcus aureus ATCC 25923.
