Abstract

Chives contain several chemical compounds serving as antibacterial agents: flavonoid, alkaloid, triterpenoid, tannin and saponin. This research aimed to find out and to prove n- hexane, ethyl acetate fractions and water fractions of ethanol 70% extracts of chives as antibacterial agent on Pseudomonas aeruginosa ATCC 27853 bacterium, to find out which one has most active antibacterial activity on Pseudomonas aeruginosa ATCC 27853 bacterium, ethanol extract or the three fractions, and to find out Minimum Inhibiting Concentration and Minimum Killing Concentration, viewed from most active fraction on Pseudomonas aeruginosa ATCC 27853 bacterium. Antibacterial activity test of ethanol extract, n-hexane, ethyl acetate and water fractions on Pseudomonas aeruginosa ATCC 27853 bacterium was used diffusion and dilution. Extract and fraction concentrations used in diffusion method were 25%, 20%, and 15%. Fraction concentrations used in dilution method were 25%, 12.5%, 6.25%, 3.13%, 1.56%, 0.78%, 0.39%, 0.19%, 0.04%. Most active fraction was tested for its chemical content used Thin Layer Chromatography. The results showed that the extract, n-hexane, ethyl acetate, and water fractions of chives has antibacterial activity against Pseudomonas aeruginosa ATCC 27853. Ethyl acetate was the most active antibacterial activity against Pseudomonas aeruginosa ATCC 27853. MIC can’t be observed and ethyl acetate has antibacterial activity with MKC 3,13% Keywords: Chives, diffusion method, dilution method, Pseudomonas aeruginosa ATCC 27853
