Abstract
Celery herbal (Apium graveolens L.) has antibacterial activity against Staphylococcus aureus. The active compounds in celery that have antibacterial activity are flavonoids, alkaloids, tannins, and saponins. Celery herbal extract is formulated into a cream preparation for easy use. Cream is a solid half preparation, in the form of an emulsion containing not less than 60% water and is intended for external use. Creams are topical preparations that are easy to clean and do not create an oily appearance when used. The aim of this research was to determine the antibacterial activity of the ethanol extract of celery herb cream against Staphylococcus aureus in vivo and to determine the effect of variations in the concentration of emulsifier Tween-span 60 on the physical quality and stability of the cream. Celery herb ethanol extract was prepared by maceration method using 70% ethanol solvent. The cream was made using a emulsifier tween-span 60 with a concentration variation of 3%, 4%, and 5%. The cream was tested for physical quality including organoleptic, homogeneity, pH, spreadability, adhesion, viscosity, and cream type. The results of the physical quality test analysis used two-way ANOVA. Observation of healing was seen from the length of time the infection cured after applying the cream, which was marked by the disappearance of erythema, no pus formation, and dry wounds on the rabbit's back skin. Observation of infection cure was analyzed using one way ANOVA. The test results showed that the ethanol extract cream of celery herb has good physical quality and stability. The variation in the concentration of the emulsifier tween-span 60 in the formula has an effect on the spreadability and viscosity of the cream, where the higher the emulsifier concentration, the higher viscosity and lower spreadability. The results showed that the ethanol extract of celery was antibacterial activity against Staphylococcus aureus which was infected on the skin of the rabbit's back. Formula 3 with a emulsifier tween-span 60 as much as 5% is the most optimal formula for curing Staphylococcus aureus infections.
