Abstract
Gangguan depresi merupakan gangguan medik serius yang melibatkan adanya gangguan pada neurotransmitter norepinefrin, serotonin dan dopamin. Depresi menyebabkan tingkah laku yang berbeda pada hewan uji, seperti sikap berdiam diri atau imobilitas, penurunan higenitas atau perilaku grooming. Depresi juga menyebabkan produksi berlebih pada kortisol, sehingga menyebabkan kadar glukosa darah tinggi. Senyawa brazilin yang dimurnikan dari kayu secang (Caesalpinia sappan L.) berpotensi sebagai antidepresan. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antidepresan ekstrak etanol kayu secang dan pengaruhnya terhadap kadar glukosa darah pada mencit yang diinduksi forced swim test. Mencit putih jantan diadaptasi selama 7 hari kemudian dibuat depresi dengan metode forced swim test selama 11 hari dengan durasi 5 menit. Dosis kayu secang yang digunakan dalam penelitian ini yaitu 10,69, 21,38 dan 42,74 mg/kgBB. Kontrol positif yang digunakan yaitu fluoxetin 0,052 mg/20 g BB mencit dan kontrol negatif yang digunakan yaitu Na CMC 0,5%. Mencit diukur imobility time dengan metode forced swim test, durasi grooming dengan uji cipratan sukrosa dan kadar glukosa darah menggunakan glukometer. Hasil penelitian menunjukkan bahwa ekstrak etanol kayu secang dosis 10,69; 21,38 dan 42,76 mg/KgBB mampu menurunkan imobility time secara signifikan berturut-turut sebesar 42,56; 44,43 dan 50,03% serta penurunan kadar glukosa darah berturut-turut sebesar 12,43; 16,84 dan 24,71%. Pada durasi grooming menunjukkan korelasi negatif dengan nilai imobility time. Kesimpulan dari penelitian ini yaitu induksi FST menyebabkan mencit depresi dengan ditandai oleh peningkatan IT, penurunan grooming dan peningkatan kadar glukosa darah. Dosis efektif untuk mengurangi depresi yaitu pada dosis 10,69 mg/KgBB, sedangkan dosis efektif untuk menurunkan kadar glukosa darah yaitu pada dosis 42,76 mg/KgBB. Depression causes different behaviors in test animals, such as silence or immobility, decreased hygiene or grooming behavior. Depression also causes an overproduction of cortisol, leading to high blood glucose levels. The brazilin compound purified from secang wood (Caesalpinia sappan L.) has the potential to be an antidepressant. The purpose of this study was to determine the antidepressant activity of secang wood ethanol extract and its effect on blood glucose levels in mice induced by forced swim tests. Male mice were adapted for 7 days and then depressed using the forced swim test method for 11 days with a duration of 5 minutes. The doses used in this study were 10.69; 21.38 and 42.74 mg/KgBB. The positive control used was fluoxetin 0.052 mg/20 gBB mice and the negative control used was 0.5% Na CMC. Mice were measured for imobility time using the forced swim test method, the duration of grooming with a sucrose splash test and blood glucose levels using a glucometer. The results showed that the ethanol extract of secang wood with a dose of 10,69; 21,38 and 42,76 mg /KgBB were able to significantly reduce time immobility by 42,56; 44,43 and 50,03% and the decrease in blood sugar levels respectively 12,43; 16,84 and 24,71%. The duration of the show shows negative immobility time values. The conclusion of this study is that the induction of FST leads to depression, increased IT, decreased treatment and increased blood glucose levels. The effective dose for reducing depression was at a dose of 10,69 mg / KgBB, while the effective dose for reducing blood glucose levels was at a dose of 4,76 mg / KgBB.
