Abstract
Fenofibrate is a fenofibric acid derivative that can treat dyslipidemia by increasing HDL-C, lowering LDL and cholesterol levels to normal levels. This study aims to determine the effect of variations in the concentration of lactose supertabs and concentrations of PVP K-30 in fenofibrate tablets on the physical quality of the tablets and their dissolution profile. This research made fenofibrate tablets with lactose supertab as filler binder and PVP K-30. The reason for this variation is to improve the flow properties and poor compressibility of fenofibrate, resulting in good tablets. This study consisted of three formulas with different concentrations, namely F1 (50%:5%), F2 (51.67%:3.33%), and F3 (53.33%:1.67%) for each lactose supertab. and PVP K-30. Characterization of the granule mass: flow time and angle of repose, while the physical quality of the tablets are: weight uniformity, content uniformity, hardness, friability, disintegration time, and tablet dissolution. This study utilized the direct compression method, then the test results were analyzed statistically using the SSPS program, the One Way Anova with a confidence level of 95%. The results of the physical quality test of the three formulas met the requirements for good tablets. Formula with various concentrations of lactose supertab and PVP K-30 (50%:5%) is the best formula that produces hardness of 4.40 ± 0.08, brittleness of 0.80 ± 0.04, and disintegration time of 7 ± 0.44. Parameters Q30 and DE30 in the dissolution test showed that all products produced different significance values in dissolution that met the requirements of the Indonesian Pharmacopoeia Edition VI.
