Yoanita, M. 2023. HISTOLOGY DIFFERENCES IN CHICKEN HEPAR TISSUE WITH CLEARING AND DEPARAFFINIZATION PROCESS MODIFICATIONS USING XYLOL, VINEGAR ACID AND KEROSENE. Bachelor of Applied Science in Medical Laboratory Technology, Program Faculty of Health, Setia Budi University. Histotechnics is a method for making histological preparations from certain specimens through a series of processes starting from fixation, dehydration, clearing, embedding, blocking, tissue sectioning, staining until preparations are ready to be observed using a microscope. Clearing aims to make the tissue structure look clearer, limpid, and transparent. Meanwhile, deparaffinization aims to remove paraffin contained in the tissue. The most common solvent used in these two processes is xylol, but it has harmful effects on health and is relatively expensive, so a substitute for xylol is needed, namely acetic acid and kerosene. This study aims to determine the difference in the histology appearance of chicken liver tissue with modification of the clearing and deparaffinization processes using xylol, vinegar acid and kerosene. This research was conducted at the Anatomical Pathology Laboratory of RSUD dr. Soeratno Gemolong uses descriptive research with an experimental research design. The sample is fresh chicken liver which is processed in a tissue processor using xylol, vinegar acid and kerosene in the process of clearing and deparaffinization. The results showed that in treatment I using xylol there was no change in the size and shape of cells/tissues, the nucleus was blue, clear, the cytoplasm was pink, homogeneous, the membrane was clear. Treatment II vinegar acetic acid, the cell nuclei were pale, unclear, vacuolated, the overall color was pale and did not contrast. Treatment III used kerosene, the cell nucleus was blue, clear, the cytoplasm was not stained pink, the overall color tended to be blue and did not contrast. It can be concluded that, there are differences in the histology appearance of chicken liver tissue with modification of the clearing and deparaffinization processes using xylol, vinegar acid and kerosene.
