Abstract
Loratadine is a kind of anti histamine drug, BCS class II which has poor solubility and dissolution in water. The alternative Fast Disintegrating Tablets (FDT) of loratadine in βcyclodextrin inclusion complex is one of the ways to solve the problem. This research aims to determine the effect of superdisintegrant combination between crospovidone and sodium starch glycolate on FDT loratadine on physical quality and dissolution profile. The inclusion complex was made by kneading method using ratio 1 : 2 molar, then characterized by Fourier-Transform Infrared (FTIR) and Differential Scanning Calorymetry (DSC). FDT was made by direct compression method with variation of crospovidone and sodium starch glycolate concentration ratio 5%:0; 3,75%:1,25%; 2,5%:2,5%; 1,25%:3,75%; 0:5%. The physical quality of FDT loratadine determined by tests that have been carried out include uniformity of weight, content, hardness, friability, disintegration time ( in vivo & in vitro ), wetting time, and dissolution. The research results obtained that the combination of superdisintegrant crospovidone and sodium starch glycolate ( 3,75%:1,25% ) has effect on the physical quality and the best drug release of FDT loratadin.
