Abstract

Avocado plants are one of the plants that can overcome the infection caused by bacteria. Part of the avocado plants that can be used as medicines are avocado seeds, avocado meat, and avocado fruits. Avocado leaves contain alkaloids, flavonoids, and saponins. This study aims to test the antibacterial activity of n-hexane fraction, ethyl acetate fraction, and water fraction of ethanolic extract of avocado leaf (Persea americana Mill) against Pseudomonas aeruginosa ATCC 27853 bacterium. Avocado leaf powder was extracted by maseration method using 70% ethanol solvent. The extracts obtained were fractionated with n-hexane, ethyl acetate and water solvents. Testing of Pseudomonas aeruginosa ATCC 27853 bacteria using diffusion method to find out the most active fraction then continued with dilution method to know Minimum Fill Concentration active fraction. The concentration used in the diffusion method was 50% and in the dilution method was used 50% concentration; 25%; 12.5%; 6.25%; 3.12%; 1.56%; 0.78%; 0.39%; 0.19%; 0.09%. The results showed that the fraction of n-hexane, ethyl acetate fraction, and water fraction of ethanol extract of avocado leaves had antibacterial activity against Pseudomonas aeruginosa ATCC 27853 at concentration 50%. Mean inhibitory diameter of n-hexane fraction, ethyl acetate fraction, and water fraction was 24.67 mm; 29.33 mm; and 14.00 mm. The most active fraction of Pseudomonas aeruginosa ATCC 27853 is an ethyl acetate fraction with a Minumum Kill Concentration of 12.5%.
