Abstract
Cancer is abnormal cells that invade surrounding tissue and spread to other organs. Bidara upas (Merremia mammosa (Lour.) Hall. f.) has been used empirically by the community for various diseases, one of them is breast cancer. This plant is one of basic tumor therapy and cancer by House of Research Jamu Hortus Medicus in Tawangmangu. This study aimed to determine cytotoxic test of ethyl acetate fraction of bidara upas tuber against breast cancer T47D cell lines and extract of bidara upas tuber against vero cells to see index selectivity. Extraction of bidara upas tuber was done by maceration with ethanol 96% and continued with liquid-liquid fractionation. Cytotoxic test was using MTT assay method with various concentrations (500; 250; 125; 62,5; 31,25; 15,63; 7,81) µg/mL. The data obtained were used to calculate Inhibitory Concentration 50 (IC50) value by linear regression. Selectivity index was obtained by comparing the IC50 value vero cells against cancer cells. The results showed that ethyl acetate fraction of bidara upas tuber had moderate cytotoxic activity against breast cancer T47D cell lines with IC50 of 32,3 µg/mL, while the ethanol extract of bidara upas tuber had not potential activity with IC50 of 165,6 µg/mL.
