Abstract

Breast cancer is the most common woman cancer in the world, and also in Indonesia. There has been no right therapies to resolve them. Cytotoxic agents from natural products is an alternative cancer therapies that are being developed. According to ethnomedicine, Bidara upas tuber (Merremia mammosa (Lour.) Hall. f.) has been used to treat cancer. The aim of this research was to determine the cytotoxic activity of extracts and aqueous fractions of bidara upas tuber on breast cancer T47D cell lines and also determined selectivity index. Material was extracted by maceration and continued with liquid-liquid fractination. Phytochemical screening of aqueous fraction was tested by thin layer chromatography for observed chemical compounds. The cytotoxic effect was carried out by using MTT assay. Cytotoxic activity was determined by IC50 value. The results showed that extracts and aqueous fraction of bidara upas tuber had not potential cytotoxic activity against T47D cell lines. The IC50 values of extract was 165,69 µg/mL and IC50 values of aqueous fraction was 13485,7 µg/mL. The selectivity index value was 4,46. Phytochemical screening showed that aqueous fraction contain phenol, alkaloid and terpenoid. Keyword: cytotoxic activity, T47D cell lines, aqueous fraction, Merremia mammosa.
