Abstract

Red gedy (Abelmochus manihot L. Medik) is the leaf that have an antioxidant and antidiabetic effects. This study aims to know the antioxidant effect of red gedy leaf by in vitro and in vivo methods. Ethanolic extract of red gedy leaf, is tested due to the antioxidant activity by in vitro with free radical DPPH with IC50 parameters and compared with the antioxidant activity of quercetin. Antioxidant tested by in vivo in this study using 30 rats of male white rat divided into 6 groups namely normal control, negative control, positive control (glibenklamid), extract of ethanol of red gedy leaves with dose of 100 mg, 200 mg, and 400 mg / kg bb. Measurements of increased activity of glutathione peroxidase enzyme in tissue rat liver it has been than induction with streptozotocin-nicotinamide, the data that obtained is analyze by One Way Anova (p<0,05) than continued by tukey test. The results of this study indicate that the extract of red geds ethanol has IC50 antioxidant activity of 133.57 ppm sehinnga classified as a moderate antioxidant. The results of this study indicate that administration of red gedy ethanol extract can increase the activity of glutathione peroxidase enzyme in hyperglycemic wistar rats. The most effective dose is a dose of 400 mg / kg BB of rat that shows the highest activity and is equivalent to positive control. Keywords : red gedy leaf, DPPH, glutathione peroxidase, antioxidant
