Abstract

Biologically, depression causes several disturbances in the neurotransmitters norepinephrine, serotonin and dopamine. One of the herbal medicines that can be used to treat depression is soursop leaves. The purpose of this study was to determine the antidepressant activity of soursop leaf extract (Annona muricata) in terms of the decreased duration of immobility and increased locomotor activity in mice. This research began with the preparation of ethanol extract of soursop leaves (Annona muricata) by maceration method, then carried out the characterization and phytochemical screening of the powder and extracts. The method used to trigger depression is the chronic mild stress (CMS) method for 14 days. Furthermore, 25 male mice were divided into 5 groups, namely the CMC negative control group, the amitriptyline positive control group, and the ethanol extract of soursop leaves doses of 125, 250, and 500 mg/KgBW were given for 10 days. The antidepressant test method can be seen from the increase in locomotor activity (central square and grooming) in mice using the open field test (OFT) method and using the tail suspension test (TST) to see the duration of immobility time. The results of this study indicate that soursop leaf extract has activity as an antidepressant as seen from locomotor activity (centra square and grooming) in the open field test method and immobility time with the tail suspension test method. The most effective dose to significantly increase the duration of central square, reduce grooming duration and immobility time in mice is 250 mg/kg BW.
