Abstract
Prevalence of depression due to COVID-19 pandemic increased sevenfold. Celery contains flavonoid, alkaloid, and terpenoid which are suspected to have potential as antidepressant. The objective of this study is to determine antidepressant potential extract and the most active fraction of celery leaves using the Forced Swimming Test (FST) method, and determine compound in the most active fraction. Celery leaves extract (CLE) obtained by maceration used 70% ethanol. Celery leaves fraction obtained by partition n-hexane, ethyl acetate, and water. This study used 32 mices divided into eight groups which is positive control (fluoxetine), negative control (Na-CMC suspension), three groups extract, and three groups fraction (n-hexane, ethyl acetate, and water fraction) of celery leaves. Variation dose of CLE was 150, 200, and 250 mg/kgBW. Chronic Mild Stress (CMS) was given as stressor induction. Parameters in this study are decrease duration of Immobility Time (IT) and grooming. Data analyzed use SPSS with One Way ANOVA 95% significance level and post hoc Tukey. The results of this study are CLE 150 mg/kgBW was the effective dose. Celery leaves fractions have antidepressant activity. Fraction of ethyl acetate 7,99 mg/kgBW was the most active fraction had antidepressant effect as same as fluoxetine 2,9 mg/kgBW. Flavonoid and alkaloid compound in the most active fraction possibility action in antidepressant effect. Key words: celery leaves, antidepressant, celery leaves fraction, Immobility Time
